The study on the removal of free hemoglobin by means of sepharose linked haptoglobin

Abstract
We intended to remove the free hemoglobin (Hb) directly from the blood circulating system by means of haptoglobin (Hp) which was capable to bind the free Hb.
For this purpose, soluble Hp was immobilized to the activated sepharose which was used as affinity adsorbent of the free Hb. These sepharose linked Hp were packed in Barrier Filter and attached to the extracorporeal circulation in rabbit. Blood flow rates kept at 15-25ml/min. These results were as follows.
1) Hb binding capacity of immobilized Hp (0.12-0.26g/g) was less than that of soluble Hp (0.60-0.65g/g). This loss might be due to the denaturation, deformation or inactivation of Hp in coupling phase.
2) In extracorporeal circulation, free Hb level in serum of experimental group was more reduced about 40mg/dl, compared to control group. Hb adsorbent capacity of sepharose linked Hp was about 100mg/40ml of wet volume.
3) During the circulation, leucocyte, thrombocyte and fibrinogen were abruptly decreased.
From these data, we concluted that immobilized Hp was effective to remove the free Hb.
