Incorporation of labeled glucose into glycogen in human young placental tissue was increased by about 70% with the addition of estradiol in vitro(f.c. 4×10-6M). Direct glycogen synthesis from glucose-6-14C was increased with estradiol and then reached the approximate equilibrium at one hour's incubation. Glyconeogenesis, incorporation of labeled carbon of glucose-6-14C into the carbons 1 to 5 of the glucose unit in a glycogen molecule, was operated in the course of incubation, and was increased with the addition of estradiol.
The activities of phosphoenolpyruvate carboxykinase and fructose-1, 6-diphosphatase were increasd in incubation with estradiol. This stimmulating effect of estradiol was suppressed with the addition of cycloheximide, and no effect with the actinomycin D.
Increases of placental glycogen synthesis by 17 β-estradiol added in vitro was due to the stimmulated formation of phosphoenolpyruvate carboxykinase and fructose-1, 6-diphosphatase.
