Although much attention has been given in recent years to the gas chromatographic analysis of carbohydrates in glycoproteins, few procedures have been successful in the quantitative analysis of them.
We established the gas chromatographic analysis of neutral and amino sugars in glycoproteins, which involves hydrolysis of glycoproteins with 2.5N trifluoroacetic acid, reduction of sugars to corresponding alcohols and trifluoroacetylation.
Moreover, analysis of sialic acid was performed by methanolysis followed by trifluoroacetylation. Finally, application of this procedure to T and H glycoprotein is described.
