Prostaglandin E1 (PGE1) was given intravenously in the dose of 0.1μg/kg/min for 10 minutes to human subjects consisting of 7 healthy controls, 7 hypertensives, 5 cases with diabetes mellitus, and 11 cases of hyperthyroidism.
1) All the cases responded with an increase in serum free fatty acid (FFA) and a fall in systemic blood pressure during and after the PGE1 infusion. There was, however, no distinct correlation between changes in FFA and blood pressure level.
2) Infusion of PGE1 in the dose of 0.04-0.08μg/kg/min for 60 minutes resulted in a nearly 3-fold increase in catecholamines excretion in urine together with an increase in FFA and a fall in blood pressure. Serum FFA and blood sugar did not change during and after the PGE1 infusion in 2 subjects pretreated with reserpine and guanethidine. The effect of PGE1 on serum FFA and blood sugar is considered to be mediated by the sympathetic nervous system.
3) There was no appreciable change in serum cholesterol and phospholipid levels in all the subjects studied. Serum triglyceride showed a remarkable decrease after PGE1 in patients with hyperthyroidism in contrast to a significant increase in healthy persons.
4) Lipoprotein lipase activity (LPA) was studied in 2 normal subjects and 3 patients with hyperthyroidism. LPA was not detected in 2 healthy subjects and 2 cases with hyperthyroidism in which plasma triglyceride did not significantly decrease during and after the PGE1 infusion. In one case of hyperthyroidism that responded to PGE1 with a fall in plasma triglyceride, LPA increased significantly during and after the infusion.
