A simple and rapid method for the determination of uric acid in human serum by high-performance liquid chromatography is described.Serum (0.1ml) was mixed with 1.0 ml of 0.4 mol/l perch-oric acid and the mixture was centrifuged at 2000×g for 10 min. The supernatant diluted twofold with 0.25mol/l disodium hydrogen phosphate was injected onto a reversed-phase ODS column.The mobile phase was 40 mmol/l sodium phosphate buffer, pH2.2, containing 2%methanol.The eluent was monitored at 284 nm.The lower detection limit for uric acid was 1μmol/l.The analytical recovery of uric acid was found to be almost 100%.The within-run precision (CV) was0.52% and the day-to-day CV was 1.1% for a serum uric acid concentration of 238 μmol/l.No interferences from ascorbic acid, hypoxanthine and xanthine were observed.The correlation between the value by the present method (y) and that by the uricase-catalase method of Kageyama (x) was good (y=0.997x -4.57, r=0.996).
