The preparation and antigenic properties of dehydroepiandrosterone sulphatebovine serum albumin (BSA) conjugate in which the hapten is linked to the carrier protein through the C-17 position are described. Antibody raised against this antigen in the rabbit posses sed a high affinity (Ka=1.43×109M-1) and excellent specificity to dehydroepiandrosterone 3-sulphate, exhibiting little or no cross-reactivities for related free and conjugated steroids with exceptions of 5-androstene-3β, 17β-diol 3-sulphate (67.5%) and 16α-hydroxydehydroepiandrosterone 3-sulphate (13.7%).
