We established a new procedure for the detection of alkaline phosphatase isoenzyme using 4-methylumbelliferyl phosphate as a substrate. The liberated 4-methylumbelliferone which has a strong fluorescence on the polyacrylamide gel was scanned. This methods is more sensitive than that using naphthol AS-MX phosphate as a substrate, and clinical application are also studied.
