Thin-layer chromatography (TLC) has proved to be a simple and rapid procedure for the separation of blood lipids1-3. As for the quantitative analysis of serum lipids by TLC only a few methods have been suggested4-7. They are based on two procedures. One is possible by measuring and comparing the sizes or areas of spots on a chromatoplate and the other is by determining the concentrations of various fractions obtained by eluting the scrappings of coating materials. As the former procedure, Purdy and Truter proposed the algebraic method to calculate the amount of materials from the spot area and Schlierf et al. applied this procedure to the quantitative analysis of human plasma lipids, in particular free fatty acids and triglycerides.
Araki has shown, after the documentation of the chromatogram, that serum lipid components could be estimated quantitatively by densitometry.
The purpose of the present study is to investigate whether TLC originally described by Zollner et al. can be used for the quantitative analysis of serum lipids applying the densitometry to a chromatoplate.
