Conduction block characteristics by extra-cellular photosensitization reaction in an electrical conduction-wire with cardiomyocyte

Abstract
To study basic parameters of arrhythmia treatment by a photosensitization reaction using talaporfin sodium, we analyzed the characteristic of acute electrical conduction (EC) block in cardiomyocyte line. Cardiomyocytes were cultivated on cover glass which has 60 μm cultivated wires. During 600 s photosensitization reaction, we detected EC change by cross correlation coefficient of Fluo-4 AM (Ca ion probe) fluorescence. The EC block probability reached 90% within 15 min from the start of the reaction with 30 mW/cm2. However, this probability decreased up to 50% after 2 h of the reaction. This result indicated that the acute EC block might be induced by gap junction closure caused by increase of Ca ion in cardiomyocytes.
