Influence on smooth muscle cells with long term stretch-fixing by heating dilatation: evaluaton model

Abstract

We have proposed novel short-term thermal angioplasty named Photo-thermo Dynamic Balloon (PTDB) for the treatment of arteriosclerosis. In our previous study, we found that smooth muscle cells (SMCs) were stretch-fixed after this treatment. To estimate the influences on stretch-fixing and heating of SMCs, we studied the activity evaluation method in the growth arrest state. SMCs were set the growth arrest state cultivated by 0.5 % low serum medium for 96 hours. To evaluate the SMCs' activity, we measured the quantity of NADH, the indispensable enzyme for aerobic respiration of cells using by WST-8 assay. SMCs were continuously stretched with 0-50 %. We found that the activity of the stretch-fixed smooth muscle cells increased compared to that of the non-stretched smooth muscle cells. We think that we might be investigated about smooth muscle cells' activity in the growth arrest state.
