We attempt to investigate biomolecule behavior in micro fluid on reaction field of open-well-type micro bioanalytical device (MBD) and influence of structural dimension and surface property of micro structures using multi-physics coupled simulation, and to design optimally the open-well-type MBD for high sensitivity and high reaction efficiency. Biomolecule behavior in micro fluid is affected on different forces occurred by fluid flow, electric double layer (EDL) on the surface of reaction field, Brown motion of biomolecule and so on, effect on biomolecule behavior in micro fluid. Therefore, it was simulated by solving coupled analysis considered with fluid dynamics, electrostatic force and diffusion by Brownian motion. By this simulation, it is possible to reveal the multi-physics in biomolecule behavior. Fluid flow of micro droplet, EDL, and biomolecule behavior are analyzed by computational fluid dynamics (CFD), Poisson-Boltzmann equation and particle tracing method, respectively. To simulate this phenomenon, the analysis software, COMSOL Multiphysics was used. The model that micro droplet falls on reaction field where there are micro structures and biomolecules between two micro structures is diffused with fluid flow was prepared, and fluid flow, voltage distribution, ion concentration distribution and biomolecule behavior were analyzed. As a result, the simulations were successful. On the results, we optimally design open-well-type MBD.
