This paper describes a quantitative evaluation of planar lipid bilayer formations by droplet split-and-contact method under motor manipulation. Planar lipid bilayers and lipid vesicles are used to investigate functions of membrane proteins and peptides. We developed the formation of the several types of planar lipid bilayers using droplet split-and-contact systems. There were limitations of efficiency and instability of the planar lipid bilayers because of its manual manipulation. In this work, to enhance the efficiency of the lipid bilayer formation, we introduced a stepping motor to the method to control parameters precisely such as rotation speed. To evaluate the proposing device, we observed lipid bilayer formation by high-speed camera and measured the currents of ion channels reconstituted into the lipid bilayers in various rotation speed. The device of the automated lipid bilayer formations with the optimized rotation speeds will facilitate the investigation of ion channel properties.
