Abstract

For drug discovery and pathological research of NAFLD using liver organoids, it is essential to measure local multi-parameter which are closely related to organoid status such as pH, temperature, and dissolved oxygen concentration inside the liver organoids to evaluate the fibrosis and inflammation of organoids. In this study, we proposed a multi-parameter measuring system equipped with a quartz culture well for fixing liver organoids, probe-type microsensors, and two-way observation system. Measuring system is installed in a clean bench and measures the force, pH, dissolved oxygen, etc. of organoids. In system, these wells which were embedded liver organoid are operated by a Robot Arm for automated transportation and sorting. Here, the effectiveness of the system was confirmed by experiments measuring the dissolved oxygen concentration inner liver organoids with different degrees of fibrosis.
