In order to monitor global gene expression changes during the nodulation process, we have performed SAGE analysis using the model legume, Lotus japonicus. In the last year's meeting, we have reported on the results obtained from comparative analysis of 85,482 and 80,233 tags from uninfected root and nodulating root. A statistical test has revealed 408 tags whose transcription is induced upon nodulation. The gene identification of tags was done using public as well as in-house EST sequences. Extension of the SAGE tags into 3' cDNAs was performed for characterization of unknown tags by use of the reverse SAGE (rSAGE) technique. As a next step toward understanding of the gene regulation system of the nodulation process, we initiated analyzing promoter regions obtained from the genomic sequence. Status of the project will be presented including computational analysis of the promoter sequences and establishment of the transgenic plants for assessing the promoter activity.
