The arachidonic acid metabolic activities and morphological changes during differentiation induced by dimethyl sulfoxide (DMSO) and retinoic acid (RA) were investigated in human promyelocytic leukemia (HL-60) cells. The product of the HL-60 cells after 5 days exposure to DMSO was only 5-lipoxygenase metabolites, and DMSO predominantly induced HL-60 cells to metamyelocytes. However, HL-60 cells treated with RA produced lipoxygenase metabolites as well as cyclooxygenase metabolites, and RA treatment led to the accumulation of banded and segmented neutrophils. It appears that cyclooxygenase metabolism takes place at later differentiation stage than lipoxygenase one.
