Simultaneous Analysis of Copper and Zinc in Bovine Serum by High Performance Liquid Chromatography

Abstract
Bovine serum samples were labeled with tetrakis (4-carboxyphenyl) porphine after wet digestion, and copper and zinc were simultaneously quantified by high performance liquid chromatography using piclic acid as an internal standard. The variation coefficients of copper and zinc were 0.6% and 1.4%, respectively. The recovery rates of both metals were within 100±5% and the detection limits of copper and zinc were 0.01μg/ml and 0.03μg/ml, respectively. The correlation coefficient between the data and those with the conventional colorimetry was more than 0.95. Using this method, the serum copper and zinc levels of 44 apparently healthy Holstein cows were determined to be 0.629 to 1.224μg/ml and 0.276 to 1.334μg/ml, respectively.
