Preparation of Manganese Porphyrin/Niosome System and Evaluation of Its Antioxidant Activity

Abstract
Superoxide anion radicals (O2) are produced by various organic activities that produce hydroxyl radicals (OH·) and peroxynitrites (ONOO-) with high radical toxicity. They are implicated in many diseases, including inflammation, aging, and cancer. Therefore, it is important to discover antioxidants prepared by manganese porphyrin/noisome system to enhance the property of long-term circulation in blood. Niosome is mainly composed of non-ionic interfacial active agents with low toxicity. Manganese 5,10,15,20-tetrakis-(N-2-methylpyridyl)porphyrin (MnT2MPyP) and manganese 5,10,15,20-tetrakis(4-dimethylsulfonyl)-porphyrin (MnT4Me2SuP) were synthesized and loaded into noisome composed of Pluronic F-88 and cholesterol in order to improve the function of nano-drug delivery system (n-DDS). We evaluated the manganese porphyrin/noisome system by examining fluorescence data, kinetic data obtained by stopped-flow method, and results of animal experiments using mice. We found that manganese porphyrin/noisome system had high stability and anti-oxidant activity with long-term circulation in blood. These results indicated that manganese porphyrin/noisome system was highly effective as n-DDS for antioxidant drugs.
