Extracellular vesicles are secreted by various types of cells and have been attracting attention as transport carriers of bioactive molecules such as proteins, nucleic acids, and lipids. Small extracellular vesicles such as exosomes are mainly derived from the endosomal system, and their research is being accelerated. On the other hand, the formation mechanism and regulators of plasma membrane–associated extracellular vesicles have remained unclear. Recently Bin–Amphiphysin–Rvs167 (BAR) domain–containing proteins have emerged as a potential factor in their formation. In this review, we introduce topics related to the regulation of plasma membrane morphology by BAR domain–containing proteins, the formation mechanism of extracellular vesicles by BAR proteins, and lipids in extracellular vesicles.
