Expression of cyclins, p53 and DNA content analysis in gloma

Objective: To apply laser scanning cytometry (LSC) analyze the different parts of human solid glioma to con trast with the immunohistochemical staining of cyclins, p53 protein in related to the cell cycle for judging the malignancy and prognosis of glioma. Methods: A total of 39 of different parts of glioma were studied by immunohistochemistry for p53, cyclin proteins and LSC DNA analysis from 13 patients. Tissue sections of specimens were smeared and stained for specific parameters and analyzed by LSC. On the other hand, surgical specimens were stained by immunohistochemystry method for cyclins and p53. Result: 1. We got the different positive rate of cyclin Bl, cyclin D1 and p53 proteins from different parts of glioma.2. LSC DNA content analysis were diffe rent with ploidy and DNA index from different parts of tumor. Conclusinos: 1. The expression of cyclin B1 and cyclin D1 in the center and edge of tumor is more high than the periphery of tumor.2. The expression of cyclin B1, D1 may related to the malignancy, prognosis of solid glioma,3. The rate of expression of p53 in primary cases are higher than recurrent cases.4. LSC detection of DNA content of glioma cases is useful for making the decision of malignancy, the area of resection of tumors and for judging malignancy, prognosis of patients depend on DNA ploidy.
