Guinea pigs were passively sensitized by antiserum to 2, 4-dinitrophenyl (DNP) containing passive cutaneous anaphylaxis (PCA) antibody. After 7 days, they were given an injection of the specific antigen through the stylomastoid foramen. In 20 out of 24 these guinea pigs, nystagmus and deviation characteristics were provoked within 15 minutes after the antigen injection, and subsided within 24 hr in most cases. None of 10 control animals displayed nystagmus or deviation. Endolymphatic hydrops was histologically demonstrated when the animals were sacrificed after periods of 12, 24 and 48 hr after the antigen challenge. It was preliminarily found that mast cells were present in connective tissue of the endolymphatic sac, and that carbon particles injected through the stylomastoid foramen could reach the connective tissue of the endolymphatic sac. We concluded that the characteristic nystagmus and deviation occurred as a result of a Type I allergic reaction.
