MN blood grouping of minute blood stains has been performed by various methods, but the results are not satisfactory. In the present experiments, various kinds of anti-M and -N sera such as non-adsorbed, adsorbed, and neutralized antisera and immunoglobulin class antibodies fractionated by DEAE-cellulose were applied to the group-specific double combination method (GSDCM). Minimum titers to induce the agglutination ranged from 1:1 to 1:2 in non-adsorbed antisera and from 1:64 to 1:128 in adsorbed ones. Double combinating potency (DCP) of non-adsorbed antisera decreased gradually according to the repetition of adsorptions before the antisera were specified. The similar phenomena were observed in IgG, IgA and IgM antibodies. Antisera adsorbed twice, of which DCP decreased little in the course of the adsorptions, were specified by the neutralization method with M and N active substance obtained by the phenol-saline extraction. The neutralized antisera gave evidently group-specific reaction on GSDCM, and the minimum titers were usually 1:8 to 1:16, which varied according to the proportion of antisera to the substance. Therefore, it might be considered that the neutralization is a suitable method to prepare the anti-M and -N sera for GSDCM.
