Abstract
A new high speed liquid chromatographic (HSLC) method for the determination of tocopherols (Toc) in serum is described. The Toc were extracted from 0.5ml of serum with 5.0ml of n-hexane after precipitation of proteins with ethanol. Four ml of the n-hexane extracts was evaporated under N_2 gas at 40℃. The residue was dissolved in 50μl of n-hexane. Twenty μl of the solution was injected into the HSLC column The eluted Toc were determined separa tely by using a fluorometer (Ex.298nm, Em.325nm). Fat-soluble substances other than Toc in serum sometimes interfere the determination by commonly used method, while they did not in this method. It was found that the proposed HSLC method was more simple and accurate than conventional methods for the separative determination of Toc in serum.
