Abstract
Aminoethyl derivative of Sephadex G-25 specifically adsorbed pyridoxal 5'-phosphate (PALP) among the six vitamin B_6 compounds under appropriate conditions. Optimum pH for the adsorption was around 9,and elution was effected by the use of acids, amino compounds or buffers of high ionic strength. The coloring of PALP adsorbed besides above facts indicated that the adsorption was mainly due to Schiff base formation between PALP and the aminoethyl side chain of Sephadex and also due to anion exchanger function of the same side chain. Isolation of PALP produced by incubation of pyridoxine with Escherichia coli cells was attempted by the use of aminoethyl-Sephadex columns, and a spectrophotometrically pure preparation of PALP was readily obtained.
