    ICH S7B及びE14ガイドラインのQ&Aがワーキンググループ(ICH E14/S7B IWG)の活動のもと最終化された。ICH S7Bガイドラインで概説されているように、in vitro IKr/hERGアッセイは、ヒトへの初回投与前に実施する再分極遅延とQT間隔延長のリスクを評価する上で重要な役割を果たす。さらに、マルチイオンチャネルアッセイ及びヒト心筋細胞アッセイは、フォローアップ試験として、QT間隔延長評価において重要な役割を果たす可能性がある。非臨床のin vitroアッセイは、臨床QTデータが利用可能な場合、開発後期段階における統合リスク評価にも貢献しうる。特に、ダブルネガティブ(in vitro IKr/hERG及びin vivo QTアッセイ)という非臨床結果は、TdPリスクをより正確に決定するための臨床の意思決定に使用し得る。非臨床試験において、心筋のイオン電流に対する薬物効果の効力に影響を与えるいくつかの要因が知られている。例えば、特定のイオン電流を誘発するために使用される電圧プロトコル、測定条件、データ採用のクライテリア及び採用されたデータの解析方法などが含まれる。従って、in vitro試験結果の再現性及び臨床所見へのトランスレーションを強化するために、いくつかの推奨されるベストプラクティスが提供されている。本発表では、ケーススタディの例を示しながらin vitroのベストプラクティスと統合リスク評価について紹介する。

    The paper summarizes the develo0ment of a two-dimensional (axisymmetric) and a three-dimensional mathematical model for electromagnetic casters of the two types used in the aluminum industry, which may soon be applied to other metals. A physical model where liquid metal is simulated by Wood's alloy is described and measurements of magnetic field, electric field, meniscus deformation and melt velocity reported. Some of the measurements are compared to the predictions of the 2D model and found to yield approximate agreement.

    Mechanism of electrode degradation in resistance spot welding (RSW) of magnesium alloy has been investigated using scanning electron microscopy (SEM) and energy-dispersive spectrum (EDS). The results show that electrode degradation experienced mainly four basic steps (i.e. magnesium alloy pickup, electrode alloying with magnesium, electrode tip face pitting, and cavitation). Magnesium alloy pickup began from the first weld as tiny drops of molten magnesium alloy were transferred from the sheet surface to the electrode tip face. A complex alloy layer (Cu2Mg and CuMg2) was formed by the molten magnesium alloy reacting with the electrode. Electrode pitting occurred by brittle fracture of alloyed region formed on the electrode tip face resulting in material removal from the electrode tip face. Once the electrode pitting started, large cavities formed by combining smaller pits. Therefore, it is favorable to periodic cleaning of the electrode tip face for extending electrode life and obtaining well appearance of joint surface.

    The ICH S7B and E14 Questions and Answers (Q&As) have been finalized under the auspice of the Implementation Working Group (February 2022, Step4). Since implementation of ICH S7B (October 2009, Step5), in vivo QT assay has been successful as a part of the core battery assays to safely bring investigational drugs to human studies, and lessons have been learned on how to best perform and report the results of in vivo studies. As there can be some variation in how the studies are performed, the “best practice” Q&As bring attention to certain considerations (species selection and study design, exposure assessment, heart rate correction method, assay sensitivity etc). The new E14 Q&As indicate that nonclinical assays can contribute to an integrated risk assessment for Torsades de Pointes (TdP) in clinical stages of development. Especially, double negative nonclinical assessments (in vitro IKr and in vivo QT assay) can be used for clinical decision-making to more accurately determine TdP risk. In this presentation, I will introduce in vivo QT best practice and integrated risk assessment, and share our case study for QT risk assessment considering the best practice Q&As.

    Chlorophyllin (Chl), a cupric salt of the chlorophyll, was used at concentrations of 12.5, 25, 50μg/ml in culture medium (Chl 12.5, Chl25, Chl150, respectively), in the presence or absence of mitomycin-C MMC (220×10-6M), in V79 cell culture, to assess the antimutagenic effects in the micronucleus assay on binucleate cells. Chlorophyllin was used in association with MMC in simultaneous treatments, pre and post treatments and simultaneous treatments after chlorophyllin had been pre-incubated with MMC for 1h. The results showed that chlorophyllin per se did not induce micronucleus formation at any of the concentrations assessed. The simultaneous treatment reduced significantly the micronucleus frequency at concentrations of 12.5 and 25μg, in 37.93 and 35.63%, respectively. At pre and post treatments, all the concentrations produced significant reductions: 45.98 Chl12.5), 43.68 (Chl25) and 28.73% (Chl50) in the pre treatment and of 36.78% (Chl 12.5), 51.72% (Chl25) and 37.93% (Chl50) in the post treatment. The same was observed in the simultaneous treatment after pre-incubation, where reduction of 48.27% (Chl 12.5), 57.47% (Chl25) and 58.62% (Chl50) were observed. These results indicated that chlorophyllin was not mutagenic in the system used and pointed to significant antimutagenic activity of the chlorophyllin. The data obtained in the simultaneous treatments suggested desmutagenic action whereas the pre and post treatments suggested bioantimutagenic activity of chlorophyllin in V79 cells.

    QT間隔延長リスクに注目した評価指針として、2009年にICH-S7Bガイドラインが発効され、in vitro試験及in vivoのQT測定の結果から、医薬品のQT間隔延長リスクについて統合的な評価が実施されている。ICH-S7B発効後、遅延整流カリウムイオンチャネルを介するカリウムイオン電流(IKr)以外のNa及びCaイオンチャネル電流阻害測定法や、iPS細胞由来の心筋細胞を用いた新規in vitro試験系、in silicoによる催不整脈予測モデルが新たに開発され、承認申請時のヒト催不整脈リスク評価の参考資料として用いられつつある。これらの現状を踏まえ、ICHでは、ワーキンググループ(ICH E14/S7B IWG)が形成され、これらの新規試験系について、in vitro及びin vivo試験のベストプラクティス、 in silico催不整脈モデルの作成/利用方法に関する原則、並びにこれらの非臨床試験成績を用いた統合的リスク評価に関するQ&Aが検討され、Step 4の段階まで進捗し、医薬品のヒト催不整脈リスク評価への利用が可能となっている。本発表では、Q&Aで示されている新規試験系が、承認申請時のヒトリスク評価へ用いられた際の審査時の視点について、本邦で最近上市された非循環器系薬剤等を例示して報告する。また、Q&Aで示されている新規試験系の適切性や結果の解釈の?妥当性について、PMDAへ相談等を行う場合における以下の留意点についても説明する。

    &Asにおいて安全性薬理試験の心血管系評価のための新たな推奨事項(ベストプラクティス:BP)が示され,被験物質の心室再分極遅延及びQT間隔延長のリスクについてBPで実施された試験成績を基に評価することが推奨されている.本研究ではhERG阻害剤であるcisapride及びdofetilideについて,BPに準じた測定条件によりhERG電流に対する影響を評価した.【方法】hERG発現HEK293細胞を用い,マニュアルパッチクランプ法にてhERG電流を測定した.測定はBPに準じた電圧プロトコル(ランプダウンパルス,0.2 Hz)及び温度(35~37°C)で行った.電極内液及び細胞外液組成,電圧プロトコルについてはCiPA推奨プロトコルに従った.ベースライン測定後,各hERG阻害剤を単回適用で15分間細胞に曝露しhERG電流測定を行った.続けて,1 µmol/LのE-4031(IKr選択的阻害剤)を曝露し,残留電流の測定を行った.各hERG阻害剤について4濃度(n=4)の評価を行い,hERG電流に対するIC50及びHill係数を算出した.【結果】cisapride及びdofetilideのIC50はそれぞれ16.1 nmol/L(7.8 ng/mL)及び5.4 nmol/L(2.4 ng/mL)であり,Hill係数は1.0及び0.86であった.両阻害剤は,これまでに報告されている従来法(ステップパルス,室温条件下,0.1 Hz等)と,同程度の hERG電流抑制を示した.また,BPに準じた条件にて測定を実施したCA Baronの報告(J Pharmacol Toxicol Methods,2022)と比較すると,cisaprideでは同等のhERG電流抑制がみられたが,dofetilideではわずかに強いhERG電流抑制がみられた.

    Agaricus blazei Murill is a mushroom largely consumed due to its medicinal properties. Effects of aqueous extract from its lineage AB97/11 in 2 fruiting body development stages (closed and opened pileus) were evaluated on chinese hamster V79 cells using cytokinesis blocking micronucleus (CBMN) and comet assays. The cells were treated at 0.15% concentration of aqueous extract prepared at different temperatures: ice-cold (4°C), room temperature (21°C) and warm (60°C). The extracts were applied in mutagenicity and antimutagenicity protocols (simultaneous, pre-incubation and continuous). The results showed that the aqueous extracts of Agaricus blazei lineage AB97/11 obtained at the 3 temperatures and both development stages did not present mutagenic or antimutagenic effect in V79 cells either in CBMN or comet assay.

    Multi-component silicate glasses containing alumina and alkaline earth metal oxides have been used for optical glasses. In these glasses competitive reactions between various kinds of network-forming and network-modifying oxides occur, so that it is effective to differentiate these reactions in terms of an acid-base concept. In this report the reactivities of alkali or alkaline earth oxide with P2O5, Al2O3, B2O3 and SiO2 are discussed based on the result of high-resolution solid state NMR. In alkali aluminoborosilicate glasses the population of four-coordinated boron atom, N4 are expressed well by r/(1-r). However, in the case of alkaline earth aluminoborosilicate glasses, the deviation from the line of r/(1-r) increases in the order of BaO<SrO<CaO<MgO, where r=([M2O] or [MO]-[Al2O3])/([M2O] or [MO]-[Al2O3]+[B2O3]). Alkaline earth oxide reacts with Al2O3 first. Subsequently, residual one reacts with B2O3 or SiO2. Weak base such as MgO reacts mainly with weak acids such as SiO2 rather than B2O3. On the other hand, a strong base such as BaO reacts mainly with B2O3. The equilibrium constant of the reaction, Si (Q3)+

    In the early days of railroading, locomotive engineers were recruited from among mechanics. It was a great recommendation if they had worked for a locomotive manufacturer. They were paid by day, in the same way as mechanics were. Since mechanic turned engineers were skilled in the two trade, they tend to be too proud to be obedient to orders.

    . introduced new methods of wage payment, such as the trip system and classification system. At the C.

    ., R. Harris introduced a new method by combining the two mentioned just above on Sept. 1, 1876 and reduced wage rates on June 10, 1877. Under the new method, engineers were graded into the four classes in terms of the length of the service which were paid accordingly. Judging from remaining company records, this was due partly to the financial pressure of the prolonged depression of the 1870s. The work can be done for less money than before, by replacing the first class men with the second and the third class men with lower wage rates.
    However, we must not ignore the long term significance of the new method, under which in order to climb up to the first class one has to be promoted step by step every year. In other words, the method was inseparably tied with the policy of promotion from within. On Nov. 20, 1884, Mr. Rhodes, the superintendent of the motive power, send a circular letter to all the master mechanics of the company instructing “Hereafter please do not employ engineers and firemen who had worked on other roads… In doing otherwise, we are likely to import the bad elements of other roads.” And by the middle of 1880s, the company had developed well a designed employment practices, by which it had attained self-sufficiency in well trained engineers.
    It goes without saying that this is a great achievement. At the same time, however, it bred discontent among engineers and their brotherhood and caused a bitter strike in 1888.

As a result, the temperature scaling law was given with the pinning parameters of m=2.30, p=0.92, and q=2.15. A factor decreases with the increase in the strain because not only superconducting bulk property changes but also the microstructure is affected.

    コナジラミ類は,トマトやキュウリ等の施設野菜に甚大な被害をもたらす農業害虫であり,農薬耐性の異なる複数の種やバイオタイプが確認されている。コナジラミ類が発する音の周波数スペクトル(周波数毎の強度分布)から,タバココナジラミのバイオタイプ B と Q2の判別が可能であることがこれまで明らかになっている。しかし,日本国内で主に確認されている種・バイオタイプ[オンシツコナジラミ,タバココナジラミ(バイオタイプ

    Usukizyme, a commercial enzyme preparation from Trichoderma viride, showed multiple chitin- degrading activities. One of these was purified to homogeneity by sequential DEAE Sepharose CL-6

    -Sepharose FF, and Sephacryl S-100 HR column chromatographies. The purified enzyme showed optimum activity at pH 3.5 and 50°–55°C and was stable in the pH range of 3.5–6.0 and up to 45°C. It showed higher activity toward chitosan-7B, a 62% deacetylated chitosan, as opposed to highly deacetylated chitosan substrates. Products of degradation of a 1% (w/v) solution of partially deacetylated chitin (PC-100) were purified on CM-Sephadex C-25 and analyzed by HPLC, exo-glycosidase digestion, and nitrous acid deamination. The enzyme was unable to split the GlcN-GlcN linkages in the substrate. It produced mainly (GlcNAc)2 and (GlcNAc)3 along with mixed oligosaccharides. When subjected to nitrous acid degradation, some of the mixed oligosaccharides produced mainly 2-deoxyglucitol, implying the presence of GlcN at the reducing end of the oligosaccharides.
