By electron cytochemistry diffuse Ca-ATPase (pH 8.5) and 5′-nucleotidase activities were observed in the chromosomes of Chinese hamster fibroblasts. Ca-ATPase activity was stronger in the kinetochores than in the chromatin and was not found in cells at interphase, early prometaphase and under colchicine block. 5′-nucleotidase activity was localized in the nuclear chromatin of interphase cells and in the chromosomes of intact dividing and colchicineblocked cells.
