Abstract
Ultrastructural observations and the histochemical methods for phosphatases (ACPase, ALPase, TPPase, G6Pase, and ATPase), dehydrogenases (SDH, LDH, and G6PDH), and fats were performed on the mammary and adenohypophyseal tissues of adult virgin, diestrous rats, within 48hr after a single administration of Sulpiride and in a week of the daily injection. A rapid release and the following production of secretory granules could be seen in the prolactin cells of the anterior pituitary gland 6-24hr after the injection. Acute hyperactivity of the mammary acini and ductuli could be detected 12-24hr after the single dosage, and a prominent secretion and a marked proliferation could be observed on the mammary gland during the daily injection. Milk congestion in some mammary acini caused finally a pressure atrophy in the corresponding epithelial cells. Both radioimmunoassay for prolactin contents in the serum and the adenohypophysis and radioautography for thymidine uptakes into the mammary and pituitary parenchyma did serve to confirm the “hypothalamic lactation” with the drug.
