Analysis of the Okinawa and Miyazaki Populations of the Common Silver-biddy Gerres oyena Using Random Amplified Polymorphic DNA (RAPD) Techniques

We analyzed the polymerase chain reaction (PCR) and the associated random amplification of polymorphic DNA (RAPD) techniques in order to study the Miyazaki and Okinawa populations of the common silver-biddy Gerres oyena (Forsskål). Fourty-six bands were identified for the common silverbiddy using 13 random primers. Nine bands were observed in all of the common silver biddies in this study, and the Okinawa and Miyazaki populations can be distinguished by using the A29-260 marker. In the inter-stock, the mean genetic similarity index was 0.858±0.060 between individual specimens from the Miyazaki population, 0.822±0.085 for the Okinawa population and 0.634±0.086 between them, respectively. In addition, a dendrogram distributed the genotypes (the Okinawa and Miyazaki populations) into different clusters. These results showed that the Okinawa and Miyazaki populations of the common silver-biddy are genetically distinct.
