Escherichia coli K-12 in the post growth after high hydrostatic pressure treatment (HPT) at 75 MPa for 30 min showed cell elongation due to the disorder of FtsZ ring formation induced by HPT. FtsZ ring is essential for cell division, and is also known as a bacterial cytoskeleton. Then, the effect of HPT on a eukaryotic cytoskeleton of Saccharomyces cerevisiae was also investigated. Actin cables disappeared after HPT (100 MPa) and were not reorganized until 3.5 h of growth after HPT, and while actin cables disappeared, budding did not start. In vitro polymerization of actin monomers was also highly sensitive to HPT. From these results, we concluded that the cytoskeletons are the most sensitive sites of microorganisms to HPT from the sterilization's standpoint of view.
