Compressibility is a novel measure of volume fluctuation of protein molecule. The adiabatic compressibility of enzyme is dramatically influenced by ligand binding and single amino acid substitution mainly via modified atomic packing or internal cavities. Mutants with large compressibility have high enzyme activity and allosteric properties. These results indicate that the local structure changes affect the overall protein dynamics to modulate the enzymatic function and that compressibility can give new insight into structure-function relationship of proteins.
