P-52 白色腐朽菌 Phanerochaete chrysosporium 由来βキシロシダーゼの機能解析

The cDNA encoding glycoside hydrolase family 3 beta-xylosidase (PcBXL3) was cloned from the basidiomycete Phanerochaete chrysosporium, and its recombinant protein was heterologously expressed in methylotrophic yeast Pichia pastoris. The deduced amino acid sequence of mature PcBXL3 showed significant similarity with known fungal GH family 3 beta-xylosidases such as beta-xylosidase from Trichoderma reesei. Recombinant PcBXL3 showed highest activity on pNP-beta-xyloside among various p-nitrophenyl (pNP) -glycosides. Similar to the other GH family 3 beta-xylosidases, PcBXL3 also showed activity on pNP-alpha-Arabinofranoside.
