Properties of red blood cells in KL-3R3 (CPDA-1) blood preservation solution were evaluated in vitro and In vivo. The KL-3R3 whole blood were collected from 10 healthy volunteers and stored for 6 weeks at 4 to 6°C.
In vitro the value of RBC count, Hb, Ht, ATP, 2-3DPG, in red cells red cell shapes and osmotic membrane fragility of red cells, and pH, electrolytes, glucose, and ammonia in the plasma were measured once a week during 6 weeks preservation. In addition, to evaluate the aggregates formation in KL-3R3 whole blood, we compared KL-3R3 whole blood with CPD whole blood on plasma-fibrinogen value during storage and trapped aggregates on transfusion filter membrane (KL-3R3: after 5 weeks, CPD: after 3 weeks). In vivo survival studies of autologous red cells after transfusion were carried out after storage for 5 weeks.
The changes in vitro were similar to data reported previously in overseas.
Plasma fibrinogen decreased during storage, but amounts of aggregates on transfusion filter was similar to those of CPD whole blood. The 24- and 48-hours post-transfusion survivals of 51Cr-labeled autologous red cells were 83.8±2.9% and 80.3±3.5% after 35 days storage (n=10), respectively.
KL-3R3 whole blood could be stored for 35 days with good quality, and allowed high levels of post-transfusion recovery.
