Glyphaea brevis (GB) is one of the medicinal plants used in folk medicine having various pharmacological and biological activities. Dinitrobenzene an industrial and environmental toxicant has been shown to affect the testes but the mechanism of action is unclear. The hypothesis of this study is to unravel the mechanism of action and to evaluate the effects of GB on DNB-induced TM4 Sertoli cells toxicity. TM4 Sertoli cells were exposed to GB (100 microgram/ml), DNB (10microgram/ml), GB was pre-treated and thereafter DNB for a duration of 24h and their effects were then examined. MTT assay, Inhibin B assay, GGT enzyme assay, TEER was investigated. Cell viability, Inhibin B levels, GGT enzyme activity and TEER decreased in DNB treated groups whereas an increased effect was seen in the GB treated groups. TM4 Sertoli cells pre-treated with GB was able to recover DNB-induced anti-proliferative effects, stimulate the activity of GGT enzyme, inhibin B levels, and tighten the BTB of the Sertoli cells monolayer. The protective effects of GB have great potential to alleviate oxidative stress, to confer protection of Sertoli cells from external insults and promote spermatogenesis.
