The drug content uniformity of tablets prepared by the direct compression method is greatly affected by the segregation of drugs during the mixing or tableting process. In this study, we investigated drug content uniformity of rapidly disintegrating tablets prepared using a surface-modified ibuprofen with light anhydrous silicic acid by the method established in our previous papers. We prepared five kinds of rapidly disintegrating tablets with ibuprofen contents ranging between 0.17% (the lowest content) to 34% (the highest content). Then we evaluated these preparations. When using a surface-modified ibuprofen, drug content uniformity in the mixing and tableting processes were greatly improved compared to that using an unmodified ibuprofen. It was found that rapidly disintegrating tablets with excellent drug content uniformity could be prepared by the method we established. Ibuprofen contents of these tablets ranged from one-tenth to two times that of the basic formula (drug content: 17%) used in our previous papers.
