The fluorometric method for the estimation of serum 11-hydroxycorticosteroid (11-OHCS) is simplified for effective Iaboratory works. Serum (0.2ml) was extracted with 2ml of methylene chloride. Pre-extraction of the serum with hexane, alkaline solution washing and water washing of the extract are ommitted. The most effective improvement is the application of a test tube-cell holder with commercial test tubes, with which the separation of organic phase from the fluorescence reagent is ommitted. The method showed good correlation with another fluorometric method and with radioimmnoassay. A technician can perform more than thirty assays within three hours.
