This paper investigates the staining process of multi-functional optical probing by using probe dyes at the same time. We measure the fluorescence spectra from the carp's ventricle stained with voltage-sensitive fluorescence dyes (Nile Blue A and WW781 respectively).
The experiment was done as following. First, a ventricle was removed from a carp and stained with a solution of Nile Blue A (0.02mg/ml) for 10min., and then it was perfused in Ringer's solution for 2min. to wash out unbounded Nile Blue A. Second, staining with WW781(0.1mg/ml) was done to the ventricle for 30min. and unbounded WW781 was washed out. The fluorescence emission spectra, which were pumped by an He-Ne laser from the ventricle, were measured in each staining and washing processes. After the first staining and washing, the fluorescence maximum was 678nm and after the second, it was 672nm. The fluorescence emission spectrum in the double stained ventricle consists of the fluorescence emission by both dyes.
