The resolution and imaging depth show trade off relationships in both ultrasound and optical imaging. Optical resolution photoacoustic microscope (OR-PAM) shows better resolution than acoustical resolution photoacoustic microscope (AR-PAM) while the imaging depth of the OR-PAM is limited. We have developed an OR-PAM for cell imaging with the lateral resolution of 646 nm and the axial resolution of 18 micron with the ultrasound transducer of 75 MHz. Classical type scanning acoustic microscopy (SAM) shows C-mode imaging. Especially, SAM with very high frequency ultrasound (approximately 1 GHz) uses burst wave thus B-mode image is not available. We have been developing quantitative SAM for both C-mode and B-mode imaging. A single ultrasonic pulse with the central frequency of 350 MHz is transmitted to obtain C-mode image with the distribution of speed of sound and B-mode image with acoustic impedance. In this symposium, the comparison between quantitative SAM and OR-PAM is introduced.
