The effect of amfenac sodium (Fenazox®) on experimentally induced toothache was investigated by applying reflex jaw opening introduced by electrical stimulation of the exposed tooth nerve of the lower canine. Mefenamic acid 30 mg/kg and dichlofenac sodium 5 mg/kg were used as the positive controls. The following results were obtained:
1) Amfenac sodium 1 mg/kg gave approximately the same analgesic effect as mefenamic acid 30 mg/kg. Dichlofenac sodium 5 mg/kg gave a better analgesic effect than either amfenac sodium 1 mg/kg or mefenamic acid 30 mg/kg.
2) Amfenac sodium 5 mg/kg gave the strongest analgesic effect among the drugs tested. A stronger analgesic effect than the maximum effects of dichlorofenac sodium 5 mg/kg and mefenamic acid 30 mg/kg was found 1.5 to 4 h after the administration of the drugs.
These results indicate that amfenac sodium might be an effective analgesic for the treatment of toothache.
