To study the differential responses of plant to the iso-osmotic conditions (-0.6MPa) of salt and drought stress, NaCl (150mM) and PEG 6000 (20%) were applied to young barley seedlings at the second leaf stage for 10 days.
Shoot fresh weight of barley was more severely reduced by NaCl (43%) compared to PEG (19%) after 10 days. Leaf water potential was slightly affected by PEG, but drastically decreased by NaCl. Malondialdehyde content was increased slightly by PEG but almost doubled by NaCl. Salt stress enhanced the activity of ascorbate peroxidase, catalase (CAT) and glutathion reductase, while only CAT activity was increased by drought stress. These results indicate that a different mechanism of barley might be involved in scavenging active oxygen species under mild iso-osmotic conditions of salt and drought stress, and that the CAT activity might be induced more promptly than other enzymes by a different subcellular environment under drought stress.
