A large-scale genome analysis of a model legume Lotus japonicus is in progress. The program includes collection of ESTs, genome sequencing, and generation of DNA markers and a high-density genetic linkage map. Up to present, a total of 93,000 5' and 3' ESTs have been accumulated from whole young plants, flowers, buds, pods, roots and nodules. Approximately 30 Mb of genomic sequences together with the gene information have already been released to the public. By using the determined sequences, more than 700 DNA markers have been generated and located onto the genetic map which is comprised of 6 linkage groups. Both material and information resources generated during the genome analysis, which include cDNA and genomic libraries, primer sequences and map locations of the DNA markers, and EST and genomic sequences, are extremely useful for acceleration of the analysis to understand the genetic system of this plant.
