Flagellin from Pseudomonas syringae pv. tabaci induces HR cell death on Arabidopsis thaliana. On the other hand, an oligopeptide, flg22 is known to induce general defense responses on variety of dicotyledonous plants. To investigate the effects between flagellin and flg22, we treated them on flg22-sensitive ecotype Col-0 and insensitive ecotype Ws-0 of A. thaliana. Although flagellin induced HR cell death, remarkable generation of active oxygen species (AOS) and callose deposition in both ecotypes, flg22 induced low level of generation of AOS and callose deposition in only Col-0. Thus no defense responses were observed in flg22-treated Ws-0. These results indicate that the signal transduction pathway leading to defense response from the perception of flagellin differs from that of flg22. Because FLS2, a gene encoding receptor for flg22, exits in Col-0 but not in Ws-0, besides FLS2, Arabidopsis has the receptor not for flg22, but for flagellin protein.
