Cowpea (Vigna unguiculata [L.] Walp.) is one of the world's most important legume plants that provide significant sources of the dietary protein and carbohydrate in tropics and subtropics. For high yield of cowpea under the heat stress, importance of proline accumulation in pollen has been suggested. Previously, the roles of two important enzymes for plant proline biosynthesis, which are P5CS and P5CR, have been well-characterized and studied. But, the study of proline transporters during plant stresses is still limited. The purpose of this experiment was to investigate the role of both proline-producing enzymes and also proline transporters in cowpea under abiotic stresses such as heat, drought or salinity stresses. Partial cDNAs encoding two proline transporters, VuProT1 and VuProT2, including their 3'-untranslated regions (3'-UTR) have been cloned in our study using degenerate PCR and 3'-RACE strategies. Southern blot analysis suggested that two copies of the proline transporters are present in cowpea.
