An efficient method to homogenize wheat leaf samples for double-antibody sandwich enzyme-linked immunosorbent assay（DAS-ELISA）was developed. Leaves were put into a safe-lock tube（2ml）containing 1ml of PBS-Tween buffer at a ratio of 1:20（W:Vol）and a stainless steel bead（5mm）．The tubes were shaken for 25min on a multi-specimen cell disruption device（Shake Master）．The supernatant was used in DAS-ELISA of Wheat yellow mosaic virus（WYMV）．The ELISA values of WYMV in samples homogenized with the Shake Master were similar to those in samples homogenized with mortar and pestle.
