抄録

An efficient method to homogenize wheat leaf samples for double-antibody sandwich enzyme-linked immunosorbent assay(DAS-ELISA)was developed. Leaves were put into a safe-lock tube(2ml)containing 1ml of PBS-Tween buffer at a ratio of 1:20(W:Vol)and a stainless steel bead(5mm).The tubes were shaken for 25min on a multi-specimen cell disruption device(Shake Master).The supernatant was used in DAS-ELISA of Wheat yellow mosaic virus(WYMV).The ELISA values of WYMV in samples homogenized with the Shake Master were similar to those in samples homogenized with mortar and pestle.
