The structure of heterophile human red cell antigen of the lipopolysaccharide from 'smooth' (S) strains of Shigella dysenteriae (EW-1) has been investigated.
Glucose, galactose, rhamnose, N-acetylglucosamine and nine oligosaccharides were isolated from this lipopolysaccharide by mild acid hydrolysis. Hemagglutination reaction of anti-Shigella dysenteriae chicken serum with human group O red cells was inhibited by the isolated galactose and the oligosaccharides containing galactosyl unit as the non-reducing end-group.
On the basis of the isolated oligosaccharides, it is proposed that the determinant of human heterophile red cell antigen may have a following structure; (galactosyl-)β-galactosyl-glucosyl-rhamnosyl- (1-3 or 6)-N-acetylglucosamine.
