Diphosphonate like EHDP are structural analogs of inorganic pyrophosphate which is a calcification inhibitor identified in human plasma and urine. It has been demonstrated from both in vitro and in vivo experiments that EHDP interferes with bone resorption and with the mineralization process. Now, in order to prevent the influence of preexisting bone, the effect of EHDP was studied on models, described by Urist and Reddi in which the formation of ectopic bone was induced, by implantation of demineralized homologous bone into muscular tissue. By implantation of demineralized bones, ectopic osteoneogenesis was induced. But mineralization was inhibited under the influence of continuous administration of high doses of EHDP.
