Carbamazepine (CBZ), one of the anticonvulsant drugs, is often used for the treatment of the trigeminal neuralgia.We also know that CBZ has various side effects as dizziness, somnolence, gastrointestinal disorders and so on.Kampo Herbal Medicine, Saiko-keishi-to(TJ-10) which is used for epilepsy is reported to have satisfactory effects on trigeminal neuralgia in the clinical examination.And we also reported that TJ-10 increased the pain threshold on mandibular area in chronic pain model produced by mental nerve ligation in rats. In this study, the effects of CBZ and TJ-10 on the chronic pain model were compared.In addition the effects of the other Herbal Medicines, Goreisan (TJ-17) and Juzen-taiho-to (TJ-48), were also examined.Male Wistar rats (BW:210-275g) were used.Under the pentobarbital anesthesia, right side mental nerve was ligated, and left side was taken sham operation.After the operation , animals were divided for five feeding groups.Group C:control, regular food. Group CBZ: CBZ contained food. Group 10:TJ-10 contained food. Group 17:TJ-17 contained food. Group 48:TJ-48 contained food.Pain thresholds and volume of food intaken were measured on Day14 and Day28 after the operation.CBZ and TJ-10 increased the pain thresholds on mandibular area significantly. TJ-17 and TJ-48 had tendency to increase the pain thresholds.Our results suggested that TJ-10 and CBZ have almost the same analgesic effects, and TJ-10 treatment might have the possibility instead of CBZ.
