In a preceding paper a presumption, from the results of growth experiments, chemical assay, and paper ionophoresis of the incubation products, was proposed that a stimulatory action of oxythiamine on yeast would be due to cleavage of the antagonist to its pyrimidine (oxypyrimidine) and thiazole moieties and utilization of the latter as a precursor in thiamine biosynthesis. This paper deals with isolation of oxypyrimidine from culture of Sacch. sake in a grown medium added with oxythiamine in order to ascertain the presumption above mentioned. UV spetrum, melting point, Rf value and elementary analytical data of the white crystal isolated from the incubation mixture using cellulose column chromatography, were identical with those of authentic sample of 2-methyl-4-hydroxy-5-hydroxymethylpyrimidine.
