In order to devise a new and universal method for determination of fat-soluble vitamins, studies on separation of fat-soluble vitamins by gel filtration were carried out and the following results were obtained. 1. It was found that gel filtration utilizing Sephadex LH-20 as the bed and chloroform as the eluting solvent were useful for separation of fat-soluble vitamins. 2. The values of distribution coefficient K_D and K_av for about 30 kinds of fat-soluble vitamins and related compounds were determined. It was observed that a linear correlation between molecular weight and logarithm of K_D was present and the compounds containing hydroxyl or carboxyl groups were retarded on elution to the corresponding compounds having approximately similar molecular weights. 3. In order to get information on the conditions for separation of fat-soluble vitamins by gel filtration using Sephadex LH-20 column in chloroform, experiments were made. Sample volume applied to the column for complete separation of the two solutes was determined. The relationship between resolution and column size was investigated by calculation of the value of peak resolution R. The influence of flow rate on the efficiency of a column was studied by the comparison of the value of height equivalent to a theoretical plate. 4. On the basis of the results obtained by the above fundamental experiments, gel filtration was applied to separation and determination of fat-soluble vitamins. It became evident that gel filtration was useful for determinations of vitamins A and E, of vitamin A in foods, of vitamin D in the presence of vitamins A and E, separations of tocopherols, of antioxidants and purification of vitamin A.
