The accumulation of biotin from linoleic acid (LeA) by a strain (P-21-6) of bacterium isolated from soil is described. The bacterium was grown at 28℃, for 3 days, with continuous shaking in a test tube containing 4 ml of a 3 % Peptone medium. After one day growth, each of fatty acids, amino acids and sulfur compounds was added to the medium. LeA was a most effective of various fatty acids on the accumulation of biotin, and Cys, Cys and FeSO_4 were also effective. The organism accumulated biotin in amount of 500 ng per ml of medium from LeA (250 μg/ml) in the presence of Cys (350 μg/ml). In addition, aeration was effective on the accumulation of extracellular biotin. Intracellular and extracellular biotin-vitamers were clarified as a biotin and biotin plus desthiobiotin, respectively. Through the results of taxonomical characteristics, strain P-21,-6 was considered to be a Bacillus sp..
