In view of pathological roles of leukotrienes a potent selective inhibitor of arachidonate 5-lipoxygenase was developed on the basis of an earlier finding that flavonoids suppressed anaphylactic reactions. 5-Hexyloxy-3',4'-dihydroxy-6,7-dimethoxyflavone 4'-disodium phosphate as a 5-lipoxygenase inhibitor inhibited in vivo production of peptidoleukotrienes in the gerbil brain subjected to ischemia and reperfusion. The monoclonal antibodies raised against 12- and 5-lipoxygenases and cyclooxygenase were utilized for development of peroxidase-linked immunoassays of these enzymes, which allowed a quantitative determination of the enzyme amount rather than the enzyme activity. The method was applied to lipoxygenase screening in various animal tissues. A clinical case of cyclooxygenase abnormality was analyzed by the enzyme immunoassay.
