Improved Extraction Method for Vitamin B_6 Derivatives from Biological Materials with Ultrasonication

Abstract

Improved extraction method in the combination use of an ultrasonicator and 1N perchloric acid as the extraction medium was proposed, where the ultrasonication was carried out at 20kHz (25 watt) for 20sec under the dark conditions in an icebath. This newly developed method has advantageous in following points. This method is time-consumming one and B_6 derivatives in the sample, in question, were well extracted, by minimizing autooxidation and degradation. Especially, PLP, which was added to bovine serum albumin (BSA), was well recovered as its form from the PLP-BSA complex. Finally, this method is applicable to a small volume of sample without the volume change, which is critical for the determination of very little amount of vitamin.
